The system generates operating pressures of up to 420 bar and pumps the mobile phase at rates of up to 50 milliliters per minute. These characteristics allow it to work with semi-preparative columns with an internal diameter of up to 30 millimeters. Gradient holdup is minimized, reducing chromatographic band spreading and increasing the purity of the fractions collected. Sample delivery is provided in two ways. The fully automated version uses a sampler with a capacity of 132 vials, each with a capacity of 2 or 6 milliliters. The sample injected is adjustable from 0.1 microliters to 5 milliliters, covering the needs of both analytical search and preparative loading. For laboratories with a limited budget, a manual version with an injector is available, allowing the injection of up to 20 milliliters per cycle. Purified fractions are collected by an automatic manifold with switching valves. The device supports several operating modes: timed collection, collection upon reaching a preset detector signal level, or collection by a mass spectrometer signal. The basic module collects up to 216 fractions in glass tubes of four sizes or in microplates. If necessary, the capacity can be expanded to 864 fractions without requiring additional bench space. The system's key technology is the fraction delay sensor. During calibration, the device measures the transit time of the chromatographic peak from the detector to the separation valve. Based on these measurements, the program automatically compensates for transport delay, ensuring that the target compound reaches the exact collection vessel determined by the collection algorithm. This feature is especially important when working with narrow peaks in high-speed gradient methods. All system components are controlled via the Agilent OpenLAB CDS ChemStation software environment. The interface is intuitive for users experienced with Agilent chromatography equipment. Each module is equipped with a built-in leak detector, which automatically shuts down the entire system if an abnormal situation is detected, enhancing safety when working with organic solvents. The configuration can be expanded in stages, from a manual system to a fully automated system with a high-performance fraction collector.
