The Thermo Scientific Vanquish Core is a modular high-performance liquid chromatography (HPLC) system designed specifically for laboratories performing routine product quality control and batch sample analysis. The system is available in three configurations: isocratic (single solvent), quaternary (four mixing channels on the low-pressure side), and binary (two channels with high-pressure mixing). The Vanquish Core pumping system operates using SmartFlow technology, which automatically compensates for changes in eluent compressibility depending on its composition and backpressure. This solution ensures flow rate and gradient accuracy throughout the analysis cycle, regardless of external factors. The binary pump version generates operating pressures up to 700 bar with an adjustable mobile phase flow rate from 0.001 to 10 ml/min. The four-channel version has a gradient holdup volume of 679 µL, while the two-channel version has a gradient holdup volume of 400 µL. The Split Sampler C and Split Sampler CT autosamplers feature SmartInject technology, ensuring precise sample dispensing and high retention time reproducibility. The standard sample module capacity is 216 samples using 1.5 mL vials. With the optional Vanquish Charger, capacity can be expanded to 8832 positions, supporting 23 plates for 96 and 384 samples. Samples are cooled by an air flow within a temperature range of 4 to 40°C, ensuring the preservation of thermolabile compounds. The Column Compartment C thermostat maintains heating within a range of 5 to 85°C with an accuracy of ±0.5°C and a stability of ±0.05°C. Two thermostat modes are available: still air and forced air, allowing you to match conditions to those used on your existing equipment. Passive preheating of the eluent entering the column minimizes temperature gradients and prevents smearing of chromatographic bands. The detector array includes diode array detectors (DAD CG and DAD HL), variable wavelength detectors (VWD), fluorescence detectors (FLD), and evaporative light scattering detectors (CAD). The DAD CG collects data at a frequency of up to 125 Hz in the 190–800 nm wavelength range, providing a noise level of less than ±6 μAU at 254 nm. The FLD C fluorescence detector is equipped with a xenon lamp with a pulse frequency of up to 300 Hz and a spectral range of excitation from 200–630 nm and emission from 265–650 nm. The CAD detector is designed for the universal determination of non-chromophoric substances that do not absorb in the UV region. All hydraulic connections are made using tool-less Viper fittings, ensuring zero dead volume and eliminating the risk of overtightening threads during column installation. Control is provided via the Chromeleon CDS software, which includes tools for quality control, auditing, and 21 CFR Part 11 compliance. An optional touchscreen display provides the operator with visual information on system status, and the Solvent Monitor module monitors solvent levels and prevents depletion during operation.
