Agilent 6400 series mass spectrometric systems are triple quadrupole liquid chromatography detectors designed for the quantitative determination of organic compounds in complex matrices. The range includes several models: from the basic 6410 and 6420 to the flagship 6490, 6495 and 6475. The devices are in demand in food quality control, environmental monitoring, clinical research, pharmaceuticals and proteomics. The key technology of the older models is the iFunnel system, which combines a two-stage ion funnel with an Agilent Jet Stream source. The design includes a six-channel capillary located between the source and the first stage of the funnel, which increases the efficiency of ion extraction and reduces the number of neutral particles entering the analyzer. The iFunnel system provides excellent sensitivity at the femtogram level and extends the dynamic range up to six orders of magnitude. The Agilent Jet Stream ionization source uses a stream of super-hot blowing gas directed at an angle to the atomizer. The blowing gas focuses the aerosol torch and creates a thermal gradient, which increases the efficiency of desolvation and increases the number of ions reaching the mass analyzer. The source supports electrospray ionization, chemical ionization at atmospheric pressure, multimode mode, as well as HPLC-Chip nanoprotective technology. The vacuum system varies depending on the model. The younger versions are equipped with a single three-stage turbomolecular pump. The older models use one pre-vacuum pump and two turbomolecular pumps. The flagship versions have two pre-vacuum pumps, which is necessary for efficient pumping of the two-stage ion funnel. The 6495B model is additionally equipped with a vacuum valve that allows capillary replacement without vacuum relief. The mass analyzer is made according to the standard triple quadrupole scheme: the first quadrupole selects precursor ions, the hexapole cell of collisions ensures their fragmentation, and the third quadrupole separates the formed fragments. The system supports multiple reaction monitoring with the ability to simultaneously track up to 4,000 transitions in a single method. The minimum registration time for a single transition is 1 millisecond. The control is carried out through the MassHunter Workstation software, which includes tools for automatic device configuration, parameter optimization, and the creation of MRM methods. The Triggered MRM function allows you to automatically start registering additional transitions to confirm identity when a target connection is detected. The 6400 series systems are compatible with Agilent 1100, 1200, 1260 and 1290 Infinity liquid chromatographs. The preparation of the installation site requires a vibration-resistant surface capable of withstanding a weight of up to 115 kilograms for the main unit, as well as a nitrogen source with a purity of at least 99.5 percent.
