The Agilent 6475 mass spectrometric system is a triple quadrupole detector for liquid chromatography, the latest evolution of the reliable LC/MS/MS line. The device is aimed at laboratories performing routine quantitative analysis of pesticides, veterinary drugs, per- and polyfluoroalkyl substances (PFAS), nitrosamines and other target compounds in complex matrices. The system is based on the Agilent Jet Stream (AJS) ionization source, which uses a stream of super-hot blowing gas directed at an angle to the atomizer. This technology increases the efficiency of desolvation and increases the number of ions reaching the mass analyzer. The source supports several ionization modes: electrospray (ESI), chemical at atmospheric pressure (APCI), multimode (MMI, simultaneous operation of ESI and APCI), as well as nanospray for low-flow applications. The mass analyzer is designed according to a triple quadrupole scheme with a curved conical impact cell, which reduces background noise and ensures a compact device layout. Quadrupoles have a hyperbolic shape and are heated to prevent contamination. The range of registered masses ranges from 5 to 3000 atomic mass units, which makes it possible to analyze both low-molecular compounds and multiply charged peptides. The detector system includes a high-energy diode with an operating voltage of up to ± 20 kilovolts, providing a wide dynamic range of more than six orders of magnitude. A key operational feature is VacShield technology, which allows maintenance of the front of the ion path (capillary replacement) without resetting the vacuum and turning off the device. This solution reduces routine maintenance time from six hours to thirty minutes. The system is equipped with intelligent Early Maintenance Feedback (EMF) functions, which monitors the status of critical components and predicts the need for maintenance. The Intelligent Reflex algorithm automatically controls the residual sample transfer and going beyond the calibration range, if necessary, starting a reanalysis with additional rinses or dilution of the sample. The device supports up to 500 MRM transitions per second with a minimum registration time of 0.5 milliseconds per transition. Dynamic Multiple Reaction Monitoring (dMRM) allows you to track up to 4,000 transitions in one method, and trigger MRM (tMRM) automatically registers additional transitions to confirm the identity of compounds in the spectral library. The control is carried out through the MassHunter Workstation software with the SWARM automatic configuration function, which uses artificial intelligence algorithms to optimize device parameters. The software complies with the requirements of 21 CFR Part 11, which is critical for accredited laboratories. The system is compatible with Agilent 1260 and 1290 Infinity II series liquid chromatographs, including two-dimensional configurations for analyzing complex samples. The device is actively used for the screening of pesticides in food products, the analysis of per- and polyfluoroalkyl substances (PFAS) in water, pharmacokinetic studies, as well as for the quantitative determination of vitamin D in blood serum.
