The Agilent 6500 Series mass spectrometric system is a hybrid instrument combining a quadrupole mass filter and a time-of-flight analyzer. The series includes several models: 6530, 6545, 6550 iFunnel, 6545XT, 6546 and 6560 with ion mobility. The equipment is in demand in metabolomics, proteomics, pharmaceutical research, as well as for screening pesticides, veterinary drugs, and toxicological research. The heart of the system is the precision mass measurement technology, providing an accuracy of less than 1 millionth and a resolution of up to 40,000. The 6550 iFunnel is equipped with a two-stage ion funnel, which increases the efficiency of ion transfer to the analyzer and provides sensitivity at the femtogram level. The flagship version of the 6560 is complemented by an ion-mobile module for separating ions by shape and charge, which adds additional dimension when analyzing complex mixtures. The ionization is based on Agilent Jet Stream technology with a stream of super-hot blowing gas directed at an angle to the atomizer. This solution increases the efficiency of desolvation and increases sensitivity by five times compared to traditional sources. The device supports several ionization modes: electrospray (ESI), chemical at atmospheric pressure (APCI), photoionization (APPI), and MALDI for individual configurations. The quadrupole mass filter selects precursor ions with an isolation window width of up to 0.4 atomic mass units. The time-of-flight analyzer has a mass range of up to 20,000 atomic mass units and a data acquisition rate of up to 50 spectra per second. The Full ion MS/MS (All Ions MS/MS) technology allows fragmentation of all ions in a wide range without pre-selection, which is useful for non-targeted screening. The dynamic range of the device in one spectrum is up to five orders of magnitude, which allows simultaneous detection of both highly concentrated components and trace impurities. The SWARM Autotune auto-tuning optimizes ion optics parameters, increasing sensitivity up to five times for small molecules. The system is managed through the MassHunter Workstation software, which provides data collection, deconvolution of chromatographic peaks, identification of compounds from spectral libraries, and quantitative analysis. The software supports working with NIST, Wiley databases and user libraries. The device is compatible with Agilent Infinity II liquid chromatographs, two-dimensional liquid chromatography (2D-LC) and supercritical fluid chromatography (SFC) systems. It is possible to use a gas chromatograph with an APCI interface.
