The Thermo Scientific Orbitrap Exploris 480 mass spectrometric system is a hybrid instrument combining a quadrupole mass filter and a new generation orbital mass analyzer. The device is aimed at laboratories performing advanced research in the fields of proteomics, biopharmaceuticals, clinical research and metabolomics. The device is based on the proven Q Exact HF-X platform and surpasses it in terms of resolution, sensitivity and mass range. The heart of the system is a high-field orbital mass analyzer, which reaches a maximum resolution of 480,000 with a mass-charge value of 200. This resolution makes it possible to confidently separate isobaric compounds and identify modified peptides at ultra-low levels. The system supports four adjustable resolution levels, which allows you to flexibly customize the device for a specific analytical task. Ion management is based on Active Ion Management technology with the advanced Ion Routing Multipole (IRM) ion leash, which effectively captures, focuses and directs ions into the analyzer. The quadrupole mass filter selects precursor ions with an isolation window width of up to 0.4 atomic mass units. Ion fragmentation is performed in the HCD (Higher Energy Collisional Dissociation) impact cell, which ensures efficient destruction of molecules to obtain detailed MS/MS spectra. The system demonstrates outstanding speed characteristics. The maximum scanning speed reaches 40 measurements per second at a resolution of 7,500 per 200 mass charge, and the frequency of full MS/MS cycles is up to 20 fragmentation spectra per second. The dynamic range in one spectrum of the orbital analyzer exceeds 5000 to one. The sensitivity of the device is characterized by a signal-to-noise ratio of 100 to one when entering 50 femtograms of reserpine in MS/MS mode and 150 to one in SIM mode. The high accuracy of mass determination is provided by the built-in Easy-IC (internal calibration) system, which maintains a mass-axis drift of less than 1 millionth of a standard deviation in 24 hours of continuous operation. With external calibration, this indicator is less than 3 millionths. The time to switch the polarity between positive and negative ionization is less than 1 second for a full cycle at 60,000 resolution. The device is available in two configurations. The standard version has a mass range from 40 to 6000 atomic mass units. The modification with the BioPharma option, installed at the factory, extends the range to 8000 atomic mass units, which is especially important for the analysis of large biomolecules, including monoclonal antibodies and whole proteins. The system is managed through software unified with other Thermo Scientific platforms, which simplifies staff training and the transfer of methods between devices. The system supports a wide range of scanning modes: standard DDA and DIA methods, MSX spectral multiplexing, BoxCar technologies for increasing dynamic range, as well as tSIM and PRM target modes for quantitative analysis. The device integrates with the FAIMS Pro differential ion mobility interface to further reduce chemical noise when working with complex matrices.
