Bio SEC-5 sorbent is spherical particles of fully porous, high-purity silica gel with a diameter of 5 micrometers. The surface of each particle is coated with a neutral hydrophilic polymer layer, which minimizes undesirable interactions between the analyzed proteins and the active centers of silica. This coating eliminates hydrophobic adsorption and electrostatic repulsions, which distort the results of exclusion chromatography. Special column packing technology ensures high pore volume, improving peak capacity and resolution. The manufacturer produces speakers with six pore size options.: 100, 150, 300, 500, 1000 and 2000 Angstroms. Each pore size is designed for a specific range of protein molecular weights. The pores of 100 Angstroms are optimal for peptides and small proteins with a mass from 0.1 to 100 kDa, 300 Angstroms for monoclonal antibodies and proteins from 5 to 1250 kDa, 1000 Angstroms for large biomolecules and virus—like particles from 50 to 7500 kDa. The column is stable in the pH range from 2.0 to 8.5, which allows the use of a wide range of buffer solutions. The upper operating temperature limit is 80 degrees Celsius. The operating pressure is limited to 240 bar, while the recommended operating pressure is 137 bar. The typical flow rate of the mobile phase for columns with a diameter of 7.8 millimeters is in the range from 0.1 to 1.25 milliliters per minute, for columns with a diameter of 4.6 millimeters — from 0.1 to 0.4 milliliters per minute. The range includes analytical columns with internal diameters of 4.6 and 7.8 millimeters and lengths from 150 to 300 millimeters, as well as preparation columns with a diameter of 21.2 millimeters for cleaning biological samples on a laboratory scale. Security columns of appropriate diameters are available to protect the analytical columns. All speakers are supplied filled with phosphate buffer.
