The Agilent InfinityLab Poroshell 120 EC-C18 chromatographic column belongs to the class of reverse-phase sorbents based on surface-porous particles with grafted octadecyl groups and complete endcapping. The device is designed for fast, highly efficient separation of acidic, neutral and basic compounds over a wide pH range. The column is recommended as a starting point in the development of techniques for high-performance liquid chromatography due to its versatility. The particle design includes a solid core with a diameter of 1.7 micrometers and a porous outer layer with a thickness of 0.5 micrometers, which creates an overall diameter of 2.7 micrometers. The short diffusion path of the analytes into the porous layer provides high separation efficiency comparable to columns on sub-2 micron fully porous particles, but at a significantly lower pressure. Particles with diameters of 1.9 and 4 micrometers are also available for various chromatographic platforms, including ultra-high performance liquid chromatography. The sorbent is made of ultrapure silica gel with a purity of more than 99.995 percent, which minimizes the undesirable interaction of basic and polar compounds with the active centers of the surface. The stationary phase is a dimethyl-n-octadecylsilane monofilm fixed to the outer porous layer of the particle. Complete endcapping using special reagents ensures maximum surface decontamination and excellent peak shape for the main compounds. The column is stable in the pH range from 2.0 to 8.0. The recommended operating temperature is 60 degrees Celsius, however, when operating in the pH range from 6 to 8, the temperature should be lowered to 40 degrees Celsius to extend the service life. The maximum operating pressure is 600 bar, which makes the column compatible with HPLC and UVLC devices. The pore size is 120 angstroms, which is suitable for separating small molecules and peptides. The specific surface area is 130 square meters per gram, carbon loading is 10 percent. The chemical selectivity of the EC-C18 phase is close to that of the fully porous Agilent ZORBAX Eclipse Plus C18 column, which makes it easy to transfer used methods. The speaker comes with a pre-programmed identification tag for improved traceability. The 2 micrometer inlet fritter design reduces the need for complex sample preparation. Protective speakers are available to extend the life of the analytical column. Applications include pharmaceutical quality control, food analysis, environmental monitoring, peptides analysis from digested proteins, and general compound screening.
